1) Reaction set-up:

DNA 10 pmol
NEBuffer 3 (NEB #B7003) (10X) 1-5 µl
Endonuclease IV 1 µl (10 units)
H2O Up to 10-50 µl

2) Incubate at 37°C for 1 hour.
