Protocol for cleavage with McrBC (M0272)

Reaction Conditions:

1X NEBuffer 2, Supplement with 200 µg/ml Recombinant Albumin and 1 mM GTP. Incubate at 37°C.

Standard Protocol:

1. Set up the following reaction on ice.
   
   

   COMPONENT	50 µl REACTION
   NEBuffer 2 (10X)	5 µl (1X)
   Recombinant Albumin (20 mg/ml)	0.5 µl  (200 µg/ml)
   GTP (100 mM)	0.5 µl (1 mM)
   Methylated DNA	1 µg
   McrBC	1 µl  (10 units)
   dH20	up to 50 μl

    
   
   
2. Incubate at 37°C for 1-2 hours.
   
   
3. Inactivate at 65°C for 20 minutes.
